The separation of platelets (PLTs) from blood components, mainly red blood cells (RBCs) and white blood cells (WBCs), has many uses in tissue engineering. However, trapping, isolating, and separating different types of biological particles, like cells, in an effective manner is challenging in biological tests. This paper describes a simple yet effective geometry for separating PLTs from WBCs and RBCs using a combination of inertial forces and dielectrophoresis (DEP) by applying a suitable AC voltage on specific locations and creating a desired electric field due to differences in conductivity and relative permittivity of cells. The inertial focusing behavior was analyzed in an omega-shaped microchannel geometry to identify the best conditions for size-based separation of cells. Non-uniform electric fields were generated by embedded electrodes to implement the DEP mechanism, enabling more precise separation based on differences in cells’ conductivity and relative permittivity. The numerical results demonstrated that the PLTs achieved 80% separation efficiency and 100% separation purity. It could be concluded that the combined effects of passive and active forces, i.e., inertial focusing and DEP, significantly enhance the performance and accuracy of PLT isolation compared to either method alone.
