چکیده

فیبروئین ابریشم، پروتئینی فیبری با خواص مکانیکی بسیار مطلوب است که توسط دسته بزرگی از جانوران از جمله کرم بومبیکس‌موری به شکل رشته‌ای ساخته می‌شود. ساختار منحصر به فرد ابریشم، تنوع فرآوری، زیست‌سازگاری، در دسترس بودن آن به شکل‌های گوناگون، امکان اعمال مهندسی ژنتیک بر روی گونه‌های مختلف، قابلیت استریل‌شدن، شیمی سطح مناسب و تخریب‌پذیری کنترل‌شده، ابریشم را در کاربرد‌های بالینی، پزشکی، آرایشی-بهداشتی و غذایی به ماده‌ای بی‌نظیر بدل کرده است. در پژوهش حاضر استحصال و بررسی پروتئین فیبروئین از تار ابریشم ‌بومبیکس‌موری با هدف کاربردهای پزشکی و بهداشتی انجام شد. خواص فیزیکی، شیمیایی و ساختاری ابریشم توسط FTIR،   XRDو SEM مورد بررسی قرار گرفت. سلول‌های استئوبلاستی G292 به منظور بررسی خواص زیستی این بیومتریال بر روی آن کشت داده شد. نمودار طیف FTIR فیبروئین استخراج شده است که پیک‌هایی را در  1-cm 1655، 1-cm 1530، 1-cm 699  و 1-cm 1239 نشان می‌دهد. فیبروئین ابریشم همچنین پس از استخراج و پس از بازیابی با متانول از نظر میزان بلورینگی با دستگاه XRD مورد بررسی قرار گرفت که ظهور پیک  در نمونه بازیابی شده نشان از تشکیل مجدد صفحات بتا داشت. از الیاف ابریشم نیز قبل و بعد از صمغ‌زدایی با SEM تصویر‌برداری شد و قطر نسبی الیاف ابریشم بین 14 تا 24 میکرومتر تعیین شد. آزمون‌های زیست‌سازگاری بیرون‌تنیMTT برای بررسی قابلیت کاربرد آن در بدن انجام شد و سازگاری سلول‌ها در مجاورت فیبروئین به اثبات رسید. استئوبلاست‌های چسبیده بر روی داربست‌ها توسط SEM مورد بررسی قرار گرفت. بررسی‌های فیزیکی و زیستی اولیه بر روی پروتئین فیبروئین سازگاربودن آن با سیستم‌های زیستی را تایید کرد. به نظر می‌رسد پروتیئن حاضر می‌تواند پیش‌ماده مناسبی برای تولید مواد مرتبط با سیستم‌های زیستی باشد.

چکیده [English]

Silk fibroin is fibrous proteins with excellent mechanical properties which are produced by wide group of animals such as Bombyx Mori. Silk fibroin with specific molecular structure can be processed into a diverse set of morphologies. Additionally, biotechnologically produced silk proteins will allow the preparation of a new generation of protein-based bio-polymeric materials with programmed properties for a wide variety of exciting medical applications. In this study, silk protein was extracted from Bombyx Mori’s cocoons and evaluated by FTIR and XRD methods. Results showed sharp amide peaks in 1655 cm-1 and 1530 cm-1 wavelength in FTIR spectrum pattern confirming existence of fibroin. SEM images of the fibers showed continuous fibers with cross-section between 14 to 24 μm. Biocompatibility tests were carried out through seeding osteoblasts cell line G292 on 2D film as well as fibers. Adhesion and proliferation of osteoblasts were investigated by MTT assay which showed no cytotoxicity. Therefore, fibroin appears to be remarkable material for prospect application in biomedicine.
